EPIGENETIC CHANGES AND miRNA EXPRESSION IN ENDOMETRIAL CANCER

Endometrial cancer is one of the most common malignancies in the female reproductive system with a tendency to increase its incidence. The most common form of endometrial cancer is an endometrioid subtype. Despite its relatively favourable prognosis, when captured at an early stage, it is useful to mention new possibilities in its diagnosis and treatment by new molecular biomarkers. miRNAs are small non-coding RNA molecules that, at the level of epigenetic mechanisms, are involved in regulating gene expression. The molecular characteristics of miRNAs allow an incomplete complementary binding to target mRNA molecules, resulting in instability of the target mRNAs. Such „mutated“ mRNAs are unable to complete their regulatory role in the process of proteosynthesis. The resulting cells lose their physiological function and are tending to increase or die in the process of apoptosis. Thus, at the epigenetic level, miRNAs play the crucial role of major regulators of gene expression in the formation or degradation of tumour cells, and their altered expression may be associated with cancer initiation, progression, and metastatic processes. Recently, many studies with miRNA types have been described whose varying levels of expression have been associated with endometrial cancer. These molecules are relatively stable and can be obtained from a blood sample, where they are present in the form of circulating exosomes. They also maintain a stable level of expression activity in histological tissues. It makes them potentially the most reliable, early and non-invasive prognostic markers for endometrial cancer. Shortly, miRNA mimics, in endometrial cancers where the level of specific types of miRNAs is reduced, could also be used for gene replacement therapy to significantly reduce the progression or even the onset of the disease.
